Abstract: We report the design and implementation of a new pupil-engineered phase function that enables simple and robust 3D localization microscopy with a ten-fold extension in depth-of-field. Applications include single-particle tracking, super resolution microscopy and lab-on-chip. We demonstrate its application to in vivo mapping of blood flow in zebrafish.
